Morphological Transformations of SARS-CoV-2 Nucleocapsid Protein Biocondensates Mediated by Antimicrobial Peptides.

Recently, the discovery of antimicrobial peptides (AMPs) as excellent candidates for overcoming antibiotic resistance has attracted significant attention. AMPs are short peptides active against bacteria, cancer cells, and viruses. It has been shown that the SARS-CoV-2 nucleocapsid protein (N-P) undergoes liquid-liquid phase separation in the presence of RNA, resulting in biocondensate formation. These biocondensates are crucial for viral replication as they concentrate the viral RNA with the host cell`s protein machinery required for viral protein expression. Thus, N-P biocondensates are promising targets to block or slow down viral RNA transcription and consequently virion assembly. We investigated the ability of three AMPs to interfere with N-P/RNA condensates. Using microscopy techniques, supported by biophysical characterization, we found that the AMP LL-III partitions into the condensate, leading to clustering. Instead, the AMP CrACP1 partitions into the droplets without affecting their morphology but reducing their dynamics. Conversely, GKY20 leads to the formation of fibrillar structures after partitioning. It can be expected that such morphological transformation severely impairs the normal functionality of the N-P droplets and thus virion assembly. These results could pave the way for the development of a new class of AMP-based antiviral agents targeting biocondensates.

Characterization of E121K mutation of D-amino acid oxidase – Insights into mechanisms leading to amyotrophic lateral sclerosis

D-amino acid oxidase (DAO) maintains the intracellular d-serine level which modulates the activity of the N-methyl-d-aspartate receptor and its dysfunction has been linked to several neurodegenerative disorders. In targeted next-generation sequencing study by our group, E121K mutation in DAO was associated with amyotrophic lateral sclerosis (ALS) in patients from India. However, variations in molecular mechanisms caused by this mutation which leads to ALS have not been studied. Hence, we carried out comparative biophysical characterization and assay studies of the wildtype- and mutant E121K-DAO. We observed that the purified E121K-DAO was inactive and exhibited a lower affinity for the FAD cofactor and benzoate inhibitor. Structural studies revealed that the E121K mutant has higher beta-sheet content, melting temperature, and oligomeric states compared to the wildtype. Kinetic study of aggregation of the variants using thioflavin-T confirmed that the E121K-DAO was more prone to aggregation. Microscopic visualization showed that the aggregation proceeds through an intermediate step involving the formation of fibrillar structures in the E121K mutant. Our results give insights into the underlying mechanisms leading to ALS pathogenesis.

Technological and sensory properties of plant-based meat analogues containing fermented sunflower protein concentrate

Sunflower press cake is a protein-rich sidestream from oil production, that is potentially suitable for human consumption, but its utilisation is hindered by the high content of chlorogenic acid (CGA), an abundant anti-nutritional factor. Here we show that processing sunflower protein concentrate (SP) by fermentation (fSP) reduces the CGA content while increasing the level of free amino acids, but impairs the formation of fibrillar structures and generates a sour taste. After fermentation of SP pH was shifted to neutral (fSP7). Meat analogues based on SP, fSP, fSP7, and pea protein isolate were prepared by high-moisture extrusion processing, which reduced CGA levels further. The texture and colour of the extrudates were both improved by the neutral pH shift. Finally, the odour, flavour and structure were assessed by a trained sensory panel. We found that fermentation followed by a neutral pH shift encouraged the formation of fibrillar structures and prevented sour taste attributes in the meat analogue. Our data will facilitate the development of sunflower protein products that are more palatable and nutritionally beneficial for humans.

Lysozyme Amyloid Fibril Structural Variability Dependence on Initial Protein Folding State.

Amyloid fibril formation is associated with several amyloidoses, including neurodegenerative Alzheimer’s or Parkinson’s diseases. The process of such fibrillar structure formation is still not fully understood, with new mechanistic insights appearing on a regular basis. This, in turn, has limited the development of potential anti-amyloid compounds, with only a handful of effective cures or treatment modalities available. One of the multiple amyloid aggregation factors that requires further examination is the ability of proteins to form multiple, structurally distinct aggregates, based on the environmental conditions. In this work, we examine how the initial folding state affects the fibrilization of lysozyme—an amyloidogenic protein, often used in protein aggregation studies. We show that there is a correlation between the initial state of the protein and the aggregate formation lag time, rate of elongation, resulting aggregate structural variability and dye-binding properties, as well as formation lag time and rate of elongation.

Polymorphism of Alpha-Synuclein Amyloid Fibrils Depends on Ionic Strength and Protein Concentration.

Protein aggregate formation is linked with multiple amyloidoses, including Alzheimer‘s and Parkinson‘s diseases. Currently, the understanding of such fibrillar structure formation and propagation is still not sufficient, the outcome of which is a lack of potent, anti-amyloid drugs. The environmental conditions used during in vitro protein aggregation assays play an important role in determining both the aggregation kinetic parameters, as well as resulting fibril structure. In the case of alpha-synuclein, ionic strength has been shown as a crucial factor in its amyloid aggregation. In this work, we examine a large sample size of alpha-synuclein aggregation reactions under thirty different ionic strength and protein concentration combinations and determine the resulting fibril structural variations using their dye-binding properties, secondary structure and morphology. We show that both ionic strength and protein concentration determine the structural variability of alpha-synuclein amyloid fibrils and that sometimes even identical conditions can result in up to four distinct types of aggregates.

Texturization of a Blend of Pea and Destarched Oat Protein Using High-Moisture Extrusion.

Grain protein fractions have great potential as ingredients that contain high amounts of valuable nutritional components. The aim of this study was to study the rheological behavior of destarched oat and pea proteins and their blends in extrusion-like conditions with a closed cavity rheometer. Additionally, the possibility of producing fibrous structures with high-moisture extrusion from a blend of destarched oat and pea protein was investigated. In the temperature sweep measurement (60–160 °C) of the destarched oat protein concentrate and pea protein isolate blend, three denaturation and polymerization sections were observed. In addition, polymerization as a function of time was recorded in the time sweep measurements. The melting temperature of grain proteins was an important factor when producing texturized structures with a high-moisture extrusion. The formation of fibrillar structures was investigated with high-moisture extrusion from the destarched oat and pea protein blend at temperatures ranging from 140 to 170 °C. The protein–protein interactions were significantly influenced in the extruded samples. This was due to a decrease in the amount of extractable protein in selective buffers. In particular, there was a decrease in non-covalent and covalent bonds due to the formation of insoluble protein complexes.

3D Mapping Reveals a Complex and Transient Interstitial Matrix During Murine Kidney Development.

The extracellular matrix (ECM) is a network of proteins and glycosaminoglycans that provides structural and biochemical cues to cells. In the kidney, the ECM is critical for nephrogenesis; however, the dynamics of ECM composition and how it relates to 3D structure during development is unknown. Using embryonic day 14.5 (E14.5), E18.5, postnatal day 3 (P3), and adult kidneys, we fractionated proteins based on differential solubilities, performed liquid chromatography-tandem mass spectrometry, and identified changes in ECM protein content (matrisome). Decellularized kidneys were stained for ECM proteins and imaged in 3D using confocal microscopy. We observed an increase in interstitial ECM that connects the stromal mesenchyme to the basement membrane (TNXB, COL6A1, COL6A2, COL6A3) between the embryo and adult, and a transient elevation of interstitial matrix proteins (COL5A2, COL12A1, COL26A1, ELN, EMID1, FBN1, LTBP4, THSD4) at perinatal time points. Basement membrane proteins critical for metanephric induction (FRAS1, FREM2) were highest in abundance in the embryo, whereas proteins necessary for integrity of the glomerular basement membrane (COL4A3, COL4A4, COL4A5, LAMB2) were more abundant in the adult. 3D visualization revealed a complex interstitial matrix that dramatically changed over development, including the perinatal formation of fibrillar structures that appear to support the medullary rays. By correlating 3D ECM spatiotemporal organization with global protein abundance, we revealed novel changes in the interstitial matrix during kidney development. This new information regarding the ECM in developing kidneys offers the potential to inform the design of regenerative scaffolds that can guide nephrogenesis in vitro.

Mechanical and Tribological Studies of SU-8 Composites Filled With UHMWPE and Gum Acacia Under Dry and Bovine Serum Albumin Lubricated Conditions

Abstract SU-8 polymer was mixed with ultrahigh-molecular weight polyethylene (UHMWPE) particles in various weight percentages (wt%) to make composites. Uniform distribution of filler material was confirmed by optical microscopy imaging. Cross-sectional scanning electron microscopy (SEM) analysis showed the formation of fibrillar structure of UHMWPE which helped bind UHMWPE particles with SU-8 matrix. Gum acacia made a coating on the UHMWPE particles and helped in binding with the matrix. The compressive Young’s modulus and strength of the composites were found to decrease with UHMWPE filler addition. The composites were tested in ball-on-disk tribometer using 12 mm diameter stainless steel (grade 304) ball under a normal load of 20 N and sliding speed of 32 mm/s. SU-8 with 25 wt% of UHMWPE showed coefficient of friction of 0.1 and specific wear-rate of 6.27 × 10−5 mm3/Nm which were 52% and 61%, respectively, lower in comparison with those of bulk UHMWPE tested in aqueous solution of bovine serum albumin (BSA). Adding 10 wt% of gum acacia to the SU-8/UHMWPE composite had beneficial effects on the wear-rate as the specific wear-rate of this hybrid composite further dropped to 4.6 × 10−5 mm3/Nm which was 71% lower than that of bulk UHMWPE.

Systematic Investigation of Insulin Fibrillation on a Chip.

A microfluidic protein aggregation device (microPAD) that allows the user to perform a series of protein incubations with various concentrations of two reagents is demonstrated. The microfluidic device consists of 64 incubation chambers to perform individual incubations of the protein at 64 specific conditions. Parallel processes of metering reagents, stepwise concentration gradient generation, and mixing are achieved simultaneously by pneumatic valves. Fibrillation of bovine insulin was selected to test the device. The effect of insulin and sodium chloride (NaCl) concentration on the formation of fibrillar structures was studied by observing the growth rate of partially folded protein, using the fluorescent marker Thioflavin-T. Moreover, dual gradients of different NaCl and hydrochloric acid (HCl) concentrations were formed, to investigate their interactive roles in the formation of insulin fibrils and spherulites. The chip-system provides a bird’s eye view on protein aggregation, including an overview of the factors that affect the process and their interactions. This microfluidic platform is potentially useful for rapid analysis of the fibrillation of proteins associated with many misfolding-based diseases, such as quantitative and qualitative studies on amyloid growth.

Study of Nanofibrils Formation of Fibroin Protein in Specific Thermal and Acidity Conditions.

Background: Amyloid fibrils are insoluble arranged aggregates of proteins that are fibrillar in structure and related to many diseases (at least 20 types of illnesses) and also create many pathologic conditions. Therefore understanding the circumstance of fibril formation is very importantObjectives: This study aims to work on fibrillar structure formation of fibroin (as a model protein)Material and Methods: In this experimental study, fibroin was extracted from bombyx mori silk cocoon, and the concentration was obtained by Bradford method. The protein was incubated in a wide range of times (0 min to 7 days) in specific acidity and thermal conditions (pH=1.6, T=70 °C). The assays of UV-vis spectroscopy with congo red, field emission scanning electron microscopy, transmission electron microscopy, atomic force microscopy and circular dichroism spectroscopy were employed to monitor the fibrillation process.Results: Fibroin assemblies were formed upon the process of aggregation and fibril formation with a variety of morphology ranging from nanoparticles to elongated fibrils.Conclusion: The results showed progressive pathway of fibril formation

Do carbon nanotubes inhibit or promote amyloid fibrils formation?

Objectives: The purpose of the work was to study the effect of carbon nanotubes on the formation of fibril structures in lysozyme at room temperature under different pH values. Materials and methods: For the preparation of the samples, hen egg-white lysozyme protein (HEWL, Fluka), as well as single-walled (SWCNT, Sigma-Aldrich) and multi-walled (MWCNT, OOO TM “Spetsmash”, Kyiv, Ukraine) carbon nanotubes were used. Used techniques: IR-Fourier Absorption Spectroscopy; confocal microscopy. Results: In this paper, the study of molecular mechanisms of interaction of lysozyme with carbon nanotubes by vibrational spectroscopy was carried out and a conformational analysis of the formed complexes was performed. It is shown that carbon nanotubes can affect the structure of lysozyme even at room temperature and normal pH values, as evidenced by conformational changes in lysozyme due to interaction with carbon nanotubes. Complexes which are formed as a result of such interaction, have characteristic features of amyloid fibrillar structures. It reveals one of possible mechanisms of carbon nanotubes cytotoxicity. On the other hand, such a technique can be introduced to obtain model amyloid fibrils for further study. Conclusion: The method of vibtarional spectroscopy has shown that carbon nanotubes can influence the structure of lysozyme, as it is shown by the conformational analysis of the absorption band Amide I. After the interaction of lysozyme with CNT, an increase in the contribution of antiparallel β-conformation in the structure of lysozyme is observed, and the contribution of the α-helix conformation is reduced, which are characteristic features in the formation of fibrillar structures. The possibility of amyloid fibril formation without the use of high temperatures at different pH values with the interaction of lysozyme and carbon nanotubes, which can be applied as a method for obtaining the model amyloid fibrils, is shown.

Morphological Features and Rheological Properties of Combined Cellulose and Polyacrylonitrile Solutions in N-Methylmorpholine-N-oxide

The combined solutions of cellulose and PAN in N-methylmorpholine-N-oxide at total polymer contents of 18 and 25% are obtained by solid-phase dissolution. The investigation of phase composition and morphology of these systems via optical methods shows that the solutions of cellulose and PAN in N-methylmorpholine-N-oxide are incompatible and form emulsions. The action of deformation on the emulsions leads to the appearance of orientational effects. If PAN predominates in droplets of the dispersed phase of cellulose solutions, this leads to the formation of fibrillar structures under shear stress, while in the case of PAN dispersion medium double emulsions are formed, in which internal droplets of the cellulose solution are most easily extended under shear. The rheological behavior of the combined systems is the direct consequence of structural-morphological transformations proceeding during deformation of the system. An analysis of the morphological and rheological features of heterophase systems reveals the concentration interval for the moldability of composite fibers from spinning emulsions.

Bulk Oriented UHMWPE/FMWCNT Films for Tribological Applications.

Bulk oriented films based on ultrahigh molecular weight polyethylene (UHMWPE) with a drawing ratio of 35 were prepared by using a low solvent concentration. Bulk oriented films were filled with fluorinated multi-walled carbon nanotubes (FMWCNTs). The structure of bulk oriented films on UHMWPE, which were manufactured at different stages of orientation, was investigated by scanning electron microscope (SEM) and differential scanning calorimetry (DSC). The addition of FMWCNTs at a concentration of 0.05 wt % in bulk oriented UHMWPE films led to an increase in the tensile strength by 10% (up to 1020 ± 23 MPa) compared to unfilled oriented films. However, the addition of FMWCNTs at a concentration of more than 0.5 wt % led to a decrease in tensile strength due to excessive accumulation of nanotubes and hindering of self-diffusion of UHMWPE macromolecules. The multiple increase in tensile strength, doubling the hardness, the formation of fibrillar structure, and the presence of carbon nanotubes led to a significant increase in tribological properties in bulk oriented films. Bulk oriented UHMWPE/1% FMWCNT films can be operated at a maximum contact pressure that is 18 times higher and exhibit a specific wear rate more than an order of magnitude and less than the traditional UHMWPE of isotropic structure. Bulk oriented UHMWPE/1% FMWCNT films have an extremely low dry coefficient of friction (COF) of 0.075 at a contact pressure of 31 MPa. The developed bulk oriented films can be used for manufacturing frictional surfaces for sliding bearings, or for acetabular cups for knee and hip endoprostheses.

Effects of water chemistry and surface contact on the toxicity of silver nanoparticles to Bacillus subtilis.

The growing use of silver nanoparticles (AgNPs) has created concerns about its potential impacts on natural microbial communities. In this study, the physicochemical properties of AgNPs and its toxicity on natural bacteria Bacillus subtilis (B. subtilis) were investigated in aqueous conditions. The characterization data showed that AgNPs highly aggregated in aqueous conditions, and the hydrodynamic diameter of AgNPs in aqueous conditions was larger than its primary size. The studied AgNPs was less toxic to B. subtilis in estuarine water as compared to that in Milli-Q water and artificial seawater, which might be due to the observed enhanced aggregation of AgNPs in estuarine water. The toxicity of AgNPs to B. subtilis was greatly reduced when their surface contact was blocked by a dialysis membrane. Scanning electron microscope images showed that exposure contact to AgNPs resulted in damage of the microbial cell wall and enhanced formation of fibrillar structures. These results suggest that particle-cell contact is largely responsible for the observed toxicity of AgNPs in B. subtilis. This study can help to understand the potential impacts of AgNPs to natural microbes, especially in the complex aquatic environments.

N-terminal Prion Protein Peptides (PrP(120–144)) Form Parallel In-register β-Sheets via Multiple Nucleation-dependent Pathways

The prion diseases are a family of fatal neurodegenerative diseases associated with the misfolding and accumulation of normal prion protein (PrPC) into its pathogenic scrapie form (PrPSc). Understanding the fundamentals of prion protein aggregation and the molecular architecture of PrPSc is key to unraveling the pathology of prion diseases. Our work investigates the early-stage aggregation of three prion protein peptides, corresponding to residues 120-144 of human (Hu), bank vole (BV), and Syrian hamster (SHa) prion protein, from disordered monomers to β-sheet-rich fibrillar structures. Using 12 μs discontinuous molecular dynamics simulations combined with the PRIME20 force field, we find that the Hu-, BV-, and SHaPrP(120-144) aggregate via multiple nucleation-dependent pathways to form U-shaped, S-shaped, and Ω-shaped protofilaments. The S-shaped HuPrP(120-144) protofilament is similar to the amyloid core structure of HuPrP(112-141) predicted by Zweckstetter. HuPrP(120-144) has a shorter aggregation lag phase than BVPrP(120-144) followed by SHaPrP(120-144), consistent with experimental findings. Two amino acid substitutions I138M and I139M retard the formation of parallel in-register β-sheet dimers during the nucleation stage by increasing side chain-side chain association and reducing side chain interaction specificity. On average, HuPrP(120-144) aggregates contain more parallel β-sheet content than those formed by BV- and SHaPrP(120-144). Deletion of the C-terminal residues 138-144 prevents formation of fibrillar structures in agreement with the experiment. This work sheds light on the amyloid core structures underlying prion strains and how I138M, I139M, and S143N affect prion protein aggregation kinetics.

Exploring the Transition of Human α-Synuclein from Native to the Fibrillar State: Insights into the Pathogenesis of Parkinson's Disease.

The etiology of Parkinson's disease involves the interplay between the environmental and genetic factors. Here in this study human α-synuclein upon exposure to 100μM pendimethalin for 12h in vitro passes through a partially folded state which proceeds to the aggregated state and terminally ends in the fibrillar phase. Variations in the ANS fluorescence intensities led to the detection of intermediate and aggregated states at 6 and 10h respectively. Far-UV CD analysis depicted significant α-helical content for intermediate state at 6h in presence of 100μM pendimethalin. Further increasing the incubation time to 12h resulted in a predominant β-sheet content which was confirmed to be fibrillar by TEM. Turbidity, Rayleigh scattering analysis, Congo red assay and ThT measurements supported the TEM data i.e. the formation of fibrillar structure of human α-synuclein upon 12h incubation. Thus, our observation could suggest a possible underlying molecular basis for Parkinson's disease. Failure mechanisms in denture adhesives

ObjectiveThe mechanical properties of bio adhesives in oral care application are expected to be critical in defining the stability and release of devices such as dentures from the oral tissue. A multiscale experimental mechanical approach is used to evaluate the performance of denture adhesive materials. MethodsThe inherent mechanical behavior of denture fixatives was examined by separating adhesive material from a representative polymethyl methacrylate (PMMA) surface using atomic force microscopy (AFM) approaches and compared to macroscopic mechanical testing. ResultsFailure of denture adhesive material was found to be critically dependent on the formation of fibrillar structures within the adhesive. Small scale mechanical testing provided evidence for the mechanical properties of the fibrillar structures formed within the adhesive in macroscopic mechanical testing and indicated the importance of the forces required to fail the adhesive at these small length scales in controlling both the maximum forces sustained by the bulk material as well as the ease of separating the adhesive from PMMA surfaces. SignificanceOur results are important in defining the performance of denture fixative materials and their control of adhesive behavior, allowing the potential to tune properties required in the adhesion and removal of dentures.

Fibrillar structures formed by covalently bound, short, β-stranded peptides on self-assembled monolayers.

The ability to maintain or reproduce biomolecular structures on inorganic substrates has the potential to impact diverse fields such as sensing and molecular electronics, as well as the study of biological self-assembly and structure-function relationships. Because the structure and self-assembly of biomolecules are exquisitely sensitive to their local chemical and electrostatic environment, the goal of reproducing or mimicking biological function in an abiological environment, including at a surface, is challenging. However, simple and well-characterized chemical modifications of prepared surfaces can be used to tune surface chemistry, structure, electrostatics, and reactivity of inorganic materials to facilitate biofunctionalization and function. Here, we describe the covalent attachment of 13-residue β-stranded peptides containing alkyne groups to a flat gold surface functionalized with an azide-terminated self-assembled monolayer through a Huisgen cycloaddition, or "click", reaction. The chemical composition and structural morphology of these surfaces were characterized using X-ray photoelectron spectroscopy, grazing incidence angle reflection-absorption infrared spectroscopy, surface circular dichroism, and atomic force microscopy. The surface-bound β-strands self-assemble into antiparallel β-sheets to form fibrillar structures 24.9 ± 1.6 nm in diameter and 2.83 ± 0.74 nm in height on the reactive surface. The results herein provide a platform for studying and controlling the self-assembly process of biomolecules into larger supermolecular structures while allowing tunable control through chemical functionalization of the surface. Interest in the mechanisms of formation of fibrillar structures has most commonly been associated with neurodegenerative diseases, such as Alzheimer's and Parkinson's, but fibrils may actually represent the thermodynamic low-energy conformation of a much larger class of peptides and proteins. The protocol developed here is an important step toward uncovering not only the factors that dictate self-assembly but also the mechanisms by which this fibrillar class of superstructures forms.

A Conventional and 2DCOS Infrared Study of Amyloid Fibril Formation

Amyloidosis is characterized by the abnormal self-assembly and deposition of proteinaceous material into insoluble ordered aggregates. There are several pathologies associated with this aggregates, known as “protein misfolding” disorders. In many proteins, when heated at high temperature and low pH, a series of structural changes resulting in the formation of fibrillar structures are produced. Insulin is a model of fibril formation that has produced a wealth of biochemical and structural data. The time-course of fibril formation can be followed by infrared spectroscopy looking at the appearance of a characteristic band in the amide I region. The kinetics is triggered by temperature at 70 °C and pH at 2.3. The infrared spectrum shows, that after a lag time (concentration-dependent), α-helix and β-turns bands decrease first whereas the random coil component increases. Later, a band at 1626 cm-1, associated with extended chains, replaces the random coil component. Infrared 2D-COS has been applied to different stages of the process. Maps have been formed at different incubation times: before random coil formation and at different stages in the random coil-fibril exchange. The synchronous maps do not give in principle much information because they are dominated by the appereance of the band due to the fibril. In order to extract such information we have developed an approach in which we take a window and move it along the perturbation (2D-moving lapse). This, together with the asynchronous maps indicate a two-step process with a first stage associated with an opening of the protein driven by destabilization of β-turns, located in the outer part of the protein, and a second part where the fibril is formed.This work was supported in part by the Spanish Ministerio de Educacion y Ciencia (grant No. BFU 2010-22103).

Formation of Vesicles Through Solvent Assisted Self-Assembly of Hydrophobic Pentapeptides: Encapsulation and pH Responsive Release of Dyes by the Vesicles (Supplementary Material)

In the biomimetic design two hydrophobic pentapetides Boc-Ile-Aib-Leu-Phe-Ala-OMe (I) and Boc-Gly-Ile-Aib-Leu-Phe-OMe (II) (Aib: α-aminoisobutyric acid) containing one Aib each are found to undergo solvent assisted self-assembly in methanol/water to form vesicular structures, which can be disrupted by simple addition of acid. The nanovesicles are found to encapsulate dye molecules that can be released by the addition of acid as confirmed by fluorescence microscopy and UV studies. The influence of solvent polarity on the morphology of the materials generated from the peptides has been examined systematically, and shows that fibrillar structures are formed in less polar chloroform/petroleum ether mixture and vesicular structures are formed in more polar methanol/water. Single crystal X-ray diffraction studies reveal that while β-sheet mediated self-assembly leads to the formation of fibrillar structures, the solvated β-sheet structure leads to the formation of vesicular structures. The results demonstrate that even hydrophobic peptides can generate vesicular structures from polar solvent which may be employed in model studies of complex biological phenomena.
